University of Copenhagen, Department of Chemistry is offering a Postdoc position for up to three years from 1 June 2020 or soon as possible thereafter.

Background and description of the research

Our lab is developing disruptive technologies to study the biophysical properties of membranes and membrane proteins on the nanoscale using fluorescence microscopy. Membrane proteins are one of the most important classes of proteins in biology comprising more than 60% of existing pharmaceutical targets. Biological membranes enable the function of membrane proteins and therefore play a key role themselves in governing a plethora of biological processes. Our work uncovers hitherto unknown nanoscale properties of membrane and membrane proteins that ultimately dictate cell behaviour, and consequently provides a path towards new generations of therapeutic agents based on entirely new biological principles.

In a breakthrough paper in Science (Veshaguri et. al., 2016) we developed a method that resolved ionic currents with a million-fold higher sensitivity than the Nobel Prize awarded method of patch clamp (atto-amperes). This allowed us to observe for the first time the function of single transporters, revealing the existence of functional heterogeneity based on hitherto unknown off-cycle states (i.e. ultra-stable inactive and leaky states with lifetimes 105-fold longer than those of states in the classical transport cycle). The dramatic consequence of this unforeseen complexity is that many fundamental mechanistic tenets that were based on macroscopic experiments of transport will have to be revised.

Now we wish to further develop the assay described above to make it applicable to a wider range of transporters, in a more relevant biological context. Currently, the most widely applied method for single transporter study is the reconstitution of the transporter into artificial membranes, namely liposomes. Thus, a protein, often not expressed in an organism to which it is endogenous, is placed in a setting which doesn’t consist of its native lipid composition. To circumvent this, we aim to generate mammalian cell-derived vesicles occupied by transporters of interest. The task of the postdoc will be to spearhead our efforts in setting up a workflow which enables us to generate these vesicles, thus broadening the range of transporters avalible for single molecule study.

Requirements:

BSc and MSc in molecular biology

PhD focusing on extracellular vesicles

Experience in breast cancer, melanoma and colon cancer research

Experience working with laboratory animals

Experience in isolating extracellular vesicles from blood plasma

Experience in extracellular vesicle isolation from patient tissue interstitium

Cryosectioning of tissue

Immunohistochemistry

Cell culture experience

Experience handling and analyzing mass spectrometry data

Experience with Size Exclusion Chromatography

Experience performing differential, rate zonal and isopycnic ultracentrifugation

Experience with a FACS Aria and FlowJo software

Experience using a ZetaView nanoparticle tracking analyzer

The starting salary is currently up to DKK 434.179 including annual supplement (+ pension up to DKK 74.244). Negotiation for salary supplement is possible.
